Aat Bioquest Screen Quest™ Fluo-8 No Wash Calcium Assay Kit | 10 Plates
Calcium flux assays are preferred methods in drug discovery for screening G protein coupled receptors (GPCR). Screen Quest™ Fluo-8 NW Calcium Assay Kit provides a homogeneous fluorescence-based assay for detecting the intracellular calcium mobilization. Cells expressing a GPCR of interest that signals through calcium are pre-loaded with our proprietary Fluo-8 NW which can cross cell membrane. Fluo-8 NW is the brightest calcium indicator available for HTS screening. Once inside the cell, the lipophilic blocking groups of Fluo-8 AM are cleaved by non-specific cell esterase, resulting in a negatively charged fluorescent dye that stays inside cells, and its fluorescence is greatly enhanced upon binding to calcium. When cells stimulated with screening compounds, the receptor signals release of intracellular calcium, which greatly increase the fluorescence of Fluo-8 NW.
Biotium XTT Cell Viability Assay Kit (1000 Assays)
The XTT Cell Viability Assay Kit provides a simple method for determining live cell numbers using standard photometric microplate readers. Determination of live cell numbers is often used to assess the rate of cell proliferation and to screen cytotoxic agents. XTT is a tetrazolium derivative. Similar to MTT, XTT measures cell viability based on the activity of mitochondrial enzymes in live cells that reduce XTT and are inactivated shortly after cell death. Unlike MTT, XTT is reduced to a highly water-soluble orange-colored product instead of the insoluble formazan from MTT, thus eliminating the solubilization step required for the MTT assay. The amount of water-soluble product generated from XTT reduction is proportional to the number of living cells in the sample and can be photometrically quantified at 475 nm. Continuous color development permits measurements at multiple time points for extended detection range.
